Prussian blue reaction involves the treatment of sections with acid solutions of ferrocyanides. Any ferric ion (+3) present in the tissue combines with the ferrocyanide and results in the formation of a bright blue pigment called Prussian blue. This is one of the most sensitive histochemical tests and will demonstrate even single granules of iron in blood cells. Small amounts of iron are found normally in spleen and bone marrow. Excessive amounts are present in hemochromatosis, with deposits found in the liver and pancreas, hemosiderosis, with deposits in the liver, spleen, and lymph nodes. Chemical. CAS 34487-61-1. Formula C19H13O5S . Na. MW 376.36. Synthetic Water soluble pH indicator used in the 6.8 yellow to 8.2 red range and frequently used in cell biology laboratories. Widely used as cell tissue culture pH marker. A small amount of phenol red added to the growth medium will have a pink-red color under normal conditions. Typically, 15 mg/l are used for cell culture. In the event of problems, waste products produced by dying cells or overgrowth of contaminants will cause a change in pH, leading to a change in indicator color. For example, a culture of relatively slowly dividing mammalian cells can be quickly overgrown by bacterial contamination. This usually results in an acidification of the medium, turning it yellow. The waste products produced by the mammalian cells themselves will slowly decrease the pH, gradually turning the solution orange and then yellow.

Cell nuclei contain most of the cells genetic material organized as multiple long linear DNA molecules in complex with a large variety of proteins such as histones to form chromosomes. Traditionally DNA stains such as DAPI and Hoechst have been used for microscopy but require fixation and cannot be used for live cell imaging.The BioTracker 488 Green Nuclear Dye is a cell-membrane permeable green fluorescent DNA dye that specifically stains nuclei in live or fixed cells. It has excellent specificity for DNA without the need for a wash step and it has low toxicity for live cell imaging.
